About the Instrument

The Agilent (formerly Varian) Cary 5000 is a high-performance, double-beam UV-Vis-NIR spectrophotometer. It measures the absorbance, transmission, and reflectance of liquids, films, and solids. It is widely used in materials science, solar cell research, and optics to evaluate material properties across a massive wavelength range.

Measurement & Analysis

  • Absorbance (A): Determines how much light is absorbed by a sample.
  • Transmittance (%T): Measures the percentage of incident light passing through the sample.
  • Reflectance (%R): Measures light reflected from solid surfaces.
  • Diffuse Reflectance: Used for powders, rough surfaces, and highly scattering materials.
  • Specular Reflectance: Used for polished or mirror-like surfaces.
  • Optical Density (OD): Evaluates highly absorbing optical components such as filters.
  • Kinetic Measurements: Monitors changes in absorbance over time.
  • Film Thickness and Optical Constants: With appropriate accessories and software, refractive index and film thickness can be determined from transmission and reflection measurements

Applications

  • Materials Science: Characterizing nanomaterials, glass, and semiconductors across the UV-visible and near-infrared spectrum.
  • Optics and Coatings: Evaluating the optical density, transmission, and antireflective properties of viewports, filters, and thin films.
  • Solar Energy: Measuring the solar reflectance and transmittance of painted surfaces and glass to assess energy efficiency and durability, adhering to standard methods like ASTM E903-20.
  • Biochemistry & Pharmaceuticals: Analyzing the optical properties of complex biological samples like DNA, proteins, and enzymes using temperature-controlled cuvette holders.
  • Advanced Chemistry: Supporting in-situ polymerization reactions and specialized spectro-electrochemical research (e.g., CO₂ reduction).
