Genomic Sequencing Division
The Genomics Sequencing Division provides sequencing services committed to advancing genomics in Jordan. The Division maintains an array of services such as diagnosis through genotyping, mutation detection, polymorphism detection, identification of DNA fragments, detection of pathogens, oligonucleotide design and synthesis, DNA extraction, and different types of DNA forensic analysis.
At the center core of PHBC, where we are equipped with different DNA Sequencer platforms, our team of analysts have decoded hundreds of thousands of gene code in the past decade representing the haplotypes of most genetic diseases in Jordan. PHBC routinely performs the following DNA tests:
- Familial Mediterranean Fever (FMF)
- Cystic Fibrosis (CF)
- Hearing Loss (Connexin 26)
- Gaucher Disease
- Methylene Tetrahydropholate Reductase (MTHFR)
- Factor V Leiden
- Factor II Deficinceies (prothrombin)
- JAK2 (Myeloproliferative Disorders MPD)
The DNA Sequencing Lab utilizes automated Sanger sequencing method and is currently equipped with two 310 Genetic Analyzers, two 16-Capillary 3130 XL Genetic Analyzers, and several instruments for routine PCR, Gel Electrophoresis and Gel Documentation. To maximize good laboratory practice and quality control, all staff and graduate students are required to prepare their work in our isolated Master Mix facility. The lab provides solid support for researchers at JUST faculties and plays effective role in sequence-based diagnosis of genetic and infectious diseases. In addition, continuous collaboration with other Divisions at PHBC has enhanced our multidisciplinary understanding of biological processes such as the field of relationships between genetic polymorphisms and drug metabolism i.e. pharmacogenomics and individualized medicine.
DNA Synthesis Lab
The DNA synthesis lab played the key role in molecular diagnostics and outbreaks investigations regardless of its capability to design up to 25 primers per day, and this capability facilitate the process of accurate molecular diagnosis, mutation detection, pathogen detection and linkage analysis. Since the molecular diagnostic tests depend mainly on the primer’s purity and specificity to the target gene or region to be investigated, the lab is well established with high quality instrumentation that serve the goals. The well established DNA synthesis lab is equipped with ASM-800 DNA synthesizer and reverse phase OPS-201 purification system that enable the synthesis of HPLC grade, high quality, oligonucleotide primers for research and diagnostics applications.
Molecular diagnosis and molecular technologies are applicable to almost all types of samples such as amniotic fluid, chronic villus samples, hair shafts and roots, fingernails, buccul swaps, cigarettes butts, blood spots, whole blood, cultured cells, bacterial cultures, and various types of tissues. So, our DNA extraction lab is well equipped with the latest approved DNA extraction protocols and standard operating procedures that ensure the highest DNA quality, concentration and purity. On the other hand, the DNA extraction lab follows the College of American Pathologists (CAP) regulations in sample’s requisition, receiving, handling and storage in order to meet the higher standards in service and maintaining the center’s DNA banking records and proper storage for samples.
The DNA Forensic Lab maintains its forensics investigations uniqueness in Jordan because of its capability to sequence the mitochondrial DNA control region, HV1 and HV2, in cases where the nuclear DNA is absent. Our lab utilizes the Short Tandem Repeats (STRs) scanning together with the mitochondrial DNA sequencing to help the government and the justice system in solving many crime cases, paternity testing, missing individual identification and raping cases.
The lab follows the highest standards for DNA typing and detection by separating samples and reagents to prevent contamination and by running the negative and positive controls each run to ensure the best performance and accuracy. It is our honor that our forensic lab is officially recognized by the court and there were much successful collaboration.
